Abstract

Antioxidantis one of the compounds that can inactivate free radical. White layer of watermelon rind (Citrullus vulgaris, Schrad) contains flavonoid, saponin and polyphenol. Flavonoids can be useful as an antioxidant. The purpose of this study was to determine the cream of white layer watermelon rindagainst free radical and determine the optimum proportion of polysorbate 80 and sorbitan 80 mixture which can produce antioxidant cream formulation from extract of white layer watermelon rindwith optimum physical properties by simplex lattice design. White layer of watermelon rind was extracted using maceration method. Diluter used was ethanol. White layer of watermelon rind extract was made in cream with various concentrations in formula, formula A (5.25% sorbitan 80: 1.75% polysorbate 80), formula B (3.5% sorbitan 80: 3.5% polysorbate 80), formula AB (1.75% sorbitan 80: 5.25% polysorbate 80). Analysis of the test results of the three physical quality test parameters were conducted by design expert 8.0.6. method for making optimum formula. Optimum formula which obtained then conducted test of antioxidant activity against DPPH measured by spectrophotometer at wavelength of 515 nm and determined IC50value. The results showed that cream ofwhite layer watermelon rind(Citrullus vulgaris, Schrad) had absorber antioxidant activity of free radical. Antioxidant activity in 87.579 ppm cream preparation form higher than in pure extract form with IC50value was 97.389 ppm. Formula optimum of polysorbate 80 and sorbitan 80 mixture from white layer watermelon rind extract was found area in accordance with desire properties were 32% polysorbate 80 with 0.67 gram and 68% sorbitan 80 with 1.42 gram.
