Abstract

Theophylline has a relatively short half-life and a narrow therapeutic index that is 10-15µg /mL. Active substances that have a narrow therapeutic index, the concentration of the active ingredient required to be carefully controlled. Theophylline in tablet dosage formulation sustained release theophylline concentrations are expected to result in a more uniform and the blood peak levels that do not fluctuate. Based on this, research on the manufacture of slow-release theophylline tablets using xanthan gum matrix and calcium sulfate. Xanthan gum can not form a gel by it self, but when combined with an ionizable agent such us calcium sulfate will form crosslinking with xanthan gum so that it will form a gel if it is in a liquid enviroment. This study is based on the simplex lattice design (SLD) with two matrix components, namely: (100% calcium sulfate), (50% calcium sulfate: 50% xanthan gum), (100% xanthan gum). Flow time, violence and DE360 as a parameter optimization. Then calculated using the Design Expert to determine the optimum formula. The results showed that theophylline sustained-release preparations can be made, and the combination of xanthan gum matrix and calcium sulfate affect the physical quality as well as the release theophylline tablets is growing more and more proportion of xanthan gum are added to improve the flow properties granule, violence and drug release tablets and combination with addition of calcium sulfate can inhibit the release of drug from the matrix. The proportion that produces slowrelease theophylline tablets with optimum physical quality is a formula containing xanthan gum and calcium sulfate 85.71% 14.29%.
