Abstract

The escargot (Achatina fulica ferr.) mucus capsule is one of preparation forms made to facilitate the users as anti-inflammation agent. Lactose and PVP is supplementary material. Lactose is the filler and PVP is the binding material. This combination is chosen to improve the flowing property and desintegration time of escargot mucus capsule. This research aimed to find out the optimum proportion of lactose-PVP combination in escargot mucus capsule using Simplex Lattice Design. Escargot mucus derived from the escargot was cleaned and fasted for 24 hours. The mucus was then dried using lactose and aerosil. The formula based on SLD with lactose (A) and PVP (B) excipients were F1: 100% (188 mg) lactose: 0%(4mg) PVP, F2: 75%% (184 mg) lactose: 25%(8mg) PVP, F3: 50% (180 mg) lactose: 50%(12mg) PVP, F4: 25% (176 mg) lactose: 75%(16mg) PVP, F5: 0% (172 mg) lactose: 100%(2mg) PVP. The preparation of escargot mucus capsule was done using dry granulation method. The granule obtained was tested for flowing time, absorbability and desintegration time. The result of the tests was used to get SLD. The equation was used to get the response of total physical granule to find out the optimum granule formula. The data on physical properties of optimum granule formula based on SLD equation with design expert software and data of optimum formula testing were analyzed statistically using SPSS and t-test at significance level of 95%. The result of t-test showed that the optimum formula was obtained from the 0% (172 mg) lactose: 100%(20 mg) PVP combination. The optimum formula of escargot mucus capsule met the physical properties test of capsule. Keywords: Escargot (Achatina fulica ferr.), capsule, lactose, PVP, Simplex Lattice Design.
