Abstract
Red fruit (Pandanus conoideus Lam.) located in Papua has been known to have a high antioxidant content. The role of antioxidants is important in neutralizing and destroying free radicals which can cause cell damage. This study was aimed to find out the antioxidant activity of subfraction of ethyl acetate fraction from methanol extract of red fruit, and to know the level of IC50 values obtained from each subfraction of ethyl acetate fraction of red fruit methanol extract by DPPH (2,2-diphenyl-1-picrylhydrazyl) method. Red fruit extraction was done by maceration using methanol solvents, suspended in distilled water, and then fractionated to produce water, ethyl acetate and hexane fractions. The ethyl acetate fraction then separated using column chromatography (subfraction). The antioxidant activity in vitro was carried out by testing the sample ability to capture 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radicals. Then DPPH absorbance readings at a wavelength of 516 nm was conducted, the result was used to calculate the percentage reduction of DPPH free radicals. The data obtained were statistically tested using SPSS 18 program of one way anova to IC50 of water, of ethyl acetate and hexane fractions of methanol extract of red fruit. The result indicated that ethyl acetate sub-subfraction of methanol extract of red fruit (Pandanus conoideus Lam) from subfraction I, subfraction II, subfraction III, subfraction IV and subfraction V had antioxidant activity in vitro. IC50 value of subfraksi I was 14.08 ± 9.14 µg / ml, subfraksi II was 8.98x10-6 ± 1.5x10-5 µg / ml, subfraksi III was 2.24 ± 1.63 µg / ml, subfraksi IV was 2.19 ± 0.56 µg / ml and subfraksi V was 2.49 ± 0.52 µg / ml while ascorbic acid comparison had IC50 value of 0.23 ± 0.1079 µg / ml.
