Abstract

Brotowali (Tinospora crispa L. Miers) is one of the medicinal plants used to treat diabetes mellitus. Obesity can lead to insulin resistance and is one of the risk factors of diabetes mellitus. This research aims to determine antidiabetic effect of combination of infusion brotowali stem with metformin in mice which is the experience insulin resistance with obesity induction. Test animals were divided into 7 groups. One group was given normal diet and six groups were given a high-fat diet for 4 weeks. Resistance insulin was tested using insulin tolerance test with insulin injection 0.75 U/kg body weight (bw) intraperitoneally. Test preparation was given for 9 days, blood glucose levels were checked on day 5 and 9 after gave test preparation. Group I as normal control, group II as negative control was given distilled water, group III was given metformin 2.6 mg/20 g bw, group IV was given a single infusion of 11.2 mg/20 g bw, group V, VI and VII were given a combination of infusion brotowali stem with metformin 8.4 mg/20 g bw:0.32 mg/20 g bw; 5.6 mg/20 g bw:0.65 mg/20 g bw; and 2.8 mg/20 g bw:0.97 mg/20 g bw. Statistical data obtained by one way ANOVA (p<0.05). The result of this research showed that all combinations of infusion of brotowali stem and metformin could decreased blood glucose levels comparable to metformin on insulin resistance mice with obesity induction. Diabetes therapy with combination of infusion of brotowali stem and metformin should be recommended for people with diabetes mellitus because dose of metformin could be decreased so that the side effects of the metformin could be reduced.
