Abstract

Antioxidants are compounds that neutralize free radicals. Pulutan leaves (Urena lobata L.) contained flavonoids, saponins, and tanin. Presence of flavonoids encouraged to conduct research aimed to know the antioxidant activity of n-hexane, ethyl acetate and water fraction of ethanol extract of leaves of pulutan (Urena lobata L.) against DPPH radical by IC50 parameter. Pulutan leaves powder were macerated by 70% ethanol. Ethanol extract was fractionated by n-hexane, ethyl acetate, and water to separate compounds based on polarity. The compounds in the fractions and extracts were analyzed by TLC. Fractions obtained were tested antioxidant activity against DPPH radical. Free radicals activity against was measured by a spectrophotometer at a wavelength of 517 nm and the IC50 was determined. Rutin was used as positive controls. The results showed that n-hexane, ethyl acetate, and water fraction and ethanol extracts had antioxidant activity with IC50 of 59,466 ppm; 21,622 ppm; 46,270 ppm; and 52,380 ppm. Ethyl acetate fraction had the highest antioxidant activity. Key words: Urena lobata L ., Antioxidants, DPPH, Ethanol extracts, The n-hexane, ethyl acetate, and water fraction.
