Abstract

Phyllanthus buxifolius Muell, Arg is a tribe of plants Euphorbiaceae is one of the group of Indonesia medicinal plants. The research objective was to determine the effectiveness of the fraction of n-hexane, ethyl acetate, and water from the ethanolic extract of seligi stems as an antibacterial against Pseudomonas aeruginosa ATCC 27853. Extraction seligi stems with percolation method using 70% ethanol solvent, followed by solvent fractination of n-hexane, ethyl acetate, and water. Antibacteril activity testing methods used in this study is the diffusion and dilution methods. Diffusion method for measuring the area of barriers to the growth of bacteria with a concentration used was 50%; 25%; and 12,5%. Dilution method (dilution tube), a dilution series with a concentration used 25%; 12,5%; 6,25%; 3,12%; 1,56%; 0,78%; 0,39%; 0,195%; 0,097%; 0,048%. The most active fractions were tested qualitatively with Thin-Layer Chromatography (TLC). Based on research results, methods of diffusion of n-hexane fraction had no inhibition at a concentration. Ethyl acetate fraction had an average of the inhibition of 20,33 mm, 15,67 mm and 15 mm at a concentration of 50%; 25%; and 12,5%. The fraction of water has an average inhibition 16,33 mm; 13,33 mm; and 8,67 mm at a concentration of 50%; 25%; and 12,5%. Dilution method of the most active fraction has a minimum concentration to kill 1,56%. The most sctive fractions of the chemical contenf of ethyl acetate were tested qualitatively with Thin-Layer Chromatography (TLC). Identification results showed a positive fraction of ethyl acetate containing polifenol, flavonoid, saponin, and tanin.
