Abstract

Diabetes is one of the biggest causes of death in the world which the number will increase in the future. One of the therapies that can be used to heal DM is inhibition agent of α-glucosidase. Brotowali (Tinospora crispa L. Miers) plant is one of the medicinal plants used to heal diabetes mellitus which the mechanism of action hasn’t known yet. This research aims to test inhibitory activity and to determine in which concentration infusion of Brotowali stem can give 50% inhibitory effect (IC50) against α-glucosidase enzyme in vitro. Inhibition test of α-glucosidase enzyme activity is done by measuring the absorbance of p-nitrophenol (yellow) from the infusion of stem Brotowali and acarbose at 405 nm wavelength using a microplate reader. Inhibition values is determined by using the IC50 values. Statistical data is obtained from the regression equation in the subsequent independent t-test test (t-test). The result showed that the infusion of brotowali had an inhibition effect to α-glukosidase enzyme through in vitro. It also showed that the infusion of brotowali and akarbose on 22,25 mg/ml and 0,94 mg/ml concentrate could inhibit 50% α-glukosidase enzyme activity.
