Abstract
The content of flavonoids and saponins in andrographis paniculata herbs, volatile oil of black cumin and ginger are potential as anti-asthma. This study aimed to determine the effect anti-asthma and to compare the effects of a single dose administration and the combination of these three plants with the number of mast cell granules in bronchial Balb/C mice as count parameter. Test animals were divided into 8 treatment groups, consisting of 4 mice. Each treatment group were given a single extract and a combination of all three plants with a total dose of 100 mg/20 gram BW mice. Positive control used was Telfast®. Negative control used was 0,5 % CMC. Eight groups were sensitized allergenic ovalbumin (OVA) intraperitoneally on days 0 and 14. Extract single and combination performed on days 1 to 27. On day 21, 23, 25, 27, carried OVA aerosol exposure. On 28th day bronchial tissue is taken and then preparations immunohistology is made using by methylene blue staining. Preparations were observed using a light microscope with a magnification of 10 x 40 to count the number of mast cell granules. Experimental data were analyzed with the KruskalWallis test followed by Mann-Whitney test. The Conclusion, Andrographis paniculata herbs extract combination dose of 25 mg, 50 mg black cumin, ginger 25 mg/20 gram BW mice are the most effective dose in reducing the number of mast cell granules.
