Abstract
Luffa acutangula (L) Roxb seed is one of traditional medicine that have antidiabetic activity. Diabetes mellitus can be treat by α-glucosidase enzyme inhibition which play a role in carbohydrate conversion into glucose. This study was aimed to determine activity by in vitro and in vivo study of α-glucosidase enzyme inhibition and antidiabetic effect in mice alloxan–induced from ethanol extract of L. acutangula (L) Roxb seed. The method used in this study were pseudosubstrat, glucose tolerance test and alloxan induction method. Alpha glucosidase would hydrolyze p-nitrophenyl- α-D-glucopyranoside substrate into p-nitrophenol (yellow) and glucose. The enzyme activity was measured based on absorbance of p-nitrophenyl yellow. Glucose tolerance test using two phases which are first phase testing of load amount which would be given to mice which consisting of 6 test groups and second phase antihyperglycemic test with 6 groups each 5 mice. Test with alloxan induction method using 11 test groups which are negative control group, acarbose group and 8 groups of dose variation. Statistical data obtained by ANOVA. The result showed that acarbose and L. acutangula (L) Roxb seed extract had activity in inhibiting α-glucosidase enzyme, acarbose IC50 was 4.45 mg/ml, and ethanol extract of L. acutangula (L) Roxb seed IC50 was 40.17 mg / ml. The in vivo test result of glucose tolerance test of ethanol extract and acarbose decrease blood glucose levels after administration of sucrose and amylum with AUC value were 46,1 (mg/dl)/hr and 39,1(mg/dl)/hr. Alloxan induction test of Luffa acutangula (L) Roxb single extract and combination doses gave greatest average of decrease percent which were 96.98 % and 97.50% with smallest AUC value 3748.6 (mg/dL/day) and 3596.4 (mg/dL/day) Keywords : Luffa acutangula (L) Roxb, ethanol extract, α-glucosidase, alloxan, antidiabetic.
