Abstract

Cancer treatment i.e. surgery, radiotherapy and chemotherapy have many side effects. Chemopreventive agent is needed to reduce the side effect and increase effectivity of theraphy. The discovery of chemopreventive agent should heed on its selectivity to reduce side effects. The selective chemopreventive agents work effectively in cancer cells and safe for normal cells. Areca catechu seed contains flavonoids, alkaloids, and tanins substances, supposed to have anticancer property. The purpose of this study was to determine the cytotoxic effect of ethanolic extract of arecha seeds against HepG2 and Vero cell lines. Areca seeds extraction is done by the method of remaceration with ethanol solvent 70%. The parameter obtained from the cytotoxic test was IC50. HepG2 cells againts the citotoxicity test is performed by the method MTT assay with the concentration of 500; 250; 125; 62,5; 31,25; 15,625; 7,813 µg/mL. Selectivity index was determind from IC50 ratio of cancer cells to normal cells. The results showed that ethanolic extract of areca seeds had a cytotoxic activity with IC50 of 95,75 µg/mL and 477,28 µg/mL on HepG2 and Vero cells respectively. It also had high selectivity on HepG2 advance liver cancer cell line with selectivity index of 4,98. It can be concluded that the ethanolic extract of areca seeds is potential to be delevoped as anticancer agent especially on liver cancer.
