Abstract

Anchovy contains protein which is useful in the process of cell regeneration, so it can be developed into a topical gel pharmaceutical dosage form for wound healing, on the other hand protein has a very large size which makes it difficult to penetrate into the skin, so this research aims to optimize of volume comparison chitosan-TPP in forming of anchovy protein nanoparticles size which were then formulated into gel preparations containing optimum concentrations of penetran enhancer DMSO to see their penetration ability through the skin. This research was carried by formulated anchovy protein concentrate into various volume of chitosan-TPP, which was divided into 5 series, i.e. each F1 without matrix, F2 1: 1, F3 1: 3, F4 1: 5 and F5 1: 7 chitosan to TPP ratio. The formed matrices then were characterized using particle size analisys and formulated into a gel preparation containing the optimum concentration of penetran enhancer DMSO, then a diffusion test is used using Franz diffusion cells to obtain the optimum gel formula that can increase the cumulative amount and penetration rate of anchovy protein through the skin. The result showed the best volume ratio for chitosan-TPP in gel preparation with optimum concentrate of penetran enhancer DMSO was 1:7 because the matrix had small particle size 687,26 nm, good polydispersity index value 0,406, matrix encapsulation efficiency 99,11%, zeta potential value +12,9 mV, high cumulative penetrated protein 1,0333 g/cm2 and penetrated rate 0.0046 μg/cm2/minute. Results of the analysis statistical tests using One Way ANOVA showed a significant effect (p <0.05) of the chitosan-TPP matrix comparison volume on the cumulative amount and penetrated rate of gel preparation of anchovy protein concentrate ex vivo.
