Abstract
Yacon leaves is a plant from Andean mountains, Peru which are known contains phenolic compounds and derivatives that have antidiabetic activity. The purpose of this study was to determine any compounds containing in the yacon leaf fraction/ sub-fraction and find out the prediction of affinity and ligand interaction patterns of these compounds to the alpha glucosidase enzyme and PPARγ receptor. This research is a laboratory experimental research using chromatography and spectroscopy methods for separation and purification and in silico with molecular docking for prediction of affinity and interaction patterns. Ethanol extract of yacon leaves was fractionated with partition liquid fraction, VLC, classic column chromatographi and HPLC then identification with 1H-NMR and 13C-NMR. The proteins test were used 2QMJ (alpha glucosidase) and 5GTN (PPARγ) in the .pdb format obtained from the Protein Data Bank. Molecular docking using the PyRx program AutoDock Tools 4.2. The results showed that the yacon leaf fraction / sub fraction contained methyl luteonin compounds (C16H12O6) based on the 1H-MNR and 13C-NMR results. The docking results obtained the bond energy values of methyl luteolin are -6.69 kcal/mol against 2QMJ and -8.45kcal/mol against 5GTN. Affinity and interaction patterns of methyl luetolin are quite good for alpha glucosidase enzymes and PPARγ receptors compared to acarbose and pioglitazone.
