Abstract
Dementia is a chronic brain disorder characterized by a decrease in memory function. The main cause of dementia is the death of nerve cells (neurons) in the part of the brain, especially in the hippocampus caused by free radicals. Soursop leaves (Anonna muricata L.) contain phenolic compounds which are considered as the main phytochemicals responsible for antioxidant activity. This study aimed to determine the ability of soursop leaf ethanol extract to improve spatial memory, reduce malondialdehyde levels, and reduce the number of pyramidal CA1 and CA2-CA3 cells that were damaged in plumbum acetate-induced mice. This research used 30 mice which were divided into 6 groups, consisting of the normal group (CMC-Na 1%), positive control group (quercetin 25 mg / KgBW), negative control group (plumbum acetate 50 mg / KgBW), and group IV, V and VI were given soursop leaves ethanol extract with doses of 100, 200 and 400 mg/kgBB respectively. The Morris water maze spatial memory test data, malondialdehyde level data and calculation of pyramidal cell counts of CA1 and CA2-CA3 damaged areas were analyzed statistically by one-way ANOVA followed by LSD Post Hoc test with a level of trust 95 %.Mice were trained for 7 days, then on the seventh day plumbum acetate 50 mg/ Kg BW was given i.p and the test preparation given simultaneously. Results displaying the effect of soursop leaf ethanol extract on spatial memory tests of mice, malondialdehyde test and calculation of the number of pyramidal cells in CA1 and CA2-CA3 areas were significantly different between the negative control group and EEDS 400 mg/kgBW (p <0.05). The conclusion of this study shows that the administration of soursop leaf ethanol extract at a dose of 400 mg/kgBW can improve spatial memory, reduce malondialdehyde levels, and reduce the number of pyramidal CA1 and CA2-CA3 cells that have been damaged in dementia mice.
