Abstract

Diabetes mellitus with complications from diabetic nephropathy is one of the most common causes of end stage renal disease in the world. Diabetic nephropathy is a microvascular complication of diabetes mellitus due to chronic hyperclycemia. Complications that occur due to damage to the smooth blood vessels in the kidney. Black cumin seeds (Nigella sativa L.) are widely used by the community as traditional medicine. This study aims to determine the antihyperglycemic activity and the effect of ethanol extract on black cumin seeds can affect serum creatinine, BUN and reduce blood glucose levels that have been induced alloxan. This study was an laboratory experimental research using a post-test only group design. The subjects of this study were 25 male Wistar rats which conditioned type 2 diabetes by alloxan induction. Rats were grouped into 5 groups: group I normal, group II negative control alloxan 150 mg/kg BW, group III positive control glibenclamide 0,45 mg/kg BW, group IV dose of ethanol extract of black cumin seed 100 mg/kg BW and group V dose of ethanol extract of black cumin seed 200 mg/kg BW. and group VI ethanol extract of black cumin seeds 400 mg / kg BW. The effects of hypoglycemia, antioxidants and diabetic nephropathy were evaluated using parameters of blood glucose, BUN and creatianin on kidney function and renal histopathology results. The results showed that ethanol extract of black cumin seeds in doses of 100, 200 and 400 mg / kg BW could reduce blood glucose, BUN and creatinine levels in kidney function and could affect the histopathology of renal organs in rats. The dose of 400 mg / kg BW is the most effective in influencing the histopathology of kidney organs. Kata Kunci: Nigella sativa L, antihyperclycemic, nepropaty, renal histophatology.
