Abstract
Hyperglycemic in the long term can cause complications of diabetic retinopathy. Diabetic retinopathy is assessed by an increase in plasma VEGF and VEGF expression due to oxidative stress, uncoupling VEGF and NO and regulation of retinal blood vessel function. One antidiabetic retinopathy is black cumin seeds (Nigela sativa L). This study aims to determine the antihyperglycemic activity of ethanol extract of black cumin seeds and determine the decrease in VEGF levels by ethanol extract of black cumin seeds. This research is an experimental laboratory study using a post test only group design. The subjects of this study were 30 male Wistar rats conditioned by type 2 DM with streptozotocin-nicotinamide induction. Rats were grouped into 6 groups: normal group I (CMC-Na), group II negative control streptozotocin 60 mg/kg bw nicotinamide 120 mg/kg bw, group III positive control glibenclamide 0.45 mg/kg bw, group IV extract dose black cumin seeds 100 mg/kg bw, group V dose of black cumin seeds 200 mg/kg bw and group VI black cumin seeds extract 400 mg/kg bw. The results showed that ethanol extracts of black cumin seeds dose of 100, 200 and 400 mg/kg bw could reduce rat blood glucose levels, VEGF levels and could regenerate eye retina damage. The most effective dose in reducing blood glucose and VEGF is a dose of 400 mg/kg bw. Keywords: Nigela sativa L, antihyperglycemic, VEGF, retinal regeneration
