Abstract
Yacon is an Andes, PERU crop which known to have antidiabetic activity. This plant consist phenolic compounds and its derivates have antidiabetic activity. The aim of this study was to determine antidiabetic activity of extracts and fractions of yacon leaves to provide antidiabetic activity which decreasing blood glucose levels and increase insulin receptor sensitivity through GLUT-4 expression in soleus muscle in treating insulin resistance. Thirty-five male Wistar rats were randomly divided into seven groups include normal control, Na-CMC, positive control, ethanol extract, water fraction of yacon leaf, n-heksane fraction and ethyl acetate fraction. We fed male Wistar rats a high fat diet with the composition of pellet (80%), lard (15%, and quail egg yolk (5%). All rats (except normal control) were given a high fat and fructose diet treatment everyday for 45 days. Fructose is given 180mg/100g BB once per day orally. Insulin sensitivity was evaluated by insulin tolerance test. Translocation pf GLUT-4 in rat muscle using immunohistochemical. The result showed that fractions and ethanol extract of yacon leaves increase blood glucose level within 21 days in rats and 10 mg/ Kg BB ethyl acetate had given the highest percentage of hypoglycemic after normal control also increase GLUT-4 protein translocation in soleus muscle by 83,22% in Diabetes type II insulin resistance.
