Abstract

Amylase enzyme is a class of hydrolase enzyme, an enzyme capable of accelerating the process of starch hydrolysis to produce simpler molecules into disaccharides and monosaccharides. Amylase enzyme-producing organisms ranging from plants, animals, to microorganisms. Microorganisms are most widely used compared to plants and animals as a source of enzymes. Amylase enzymes are used, among others, in the food industry such as sweetener, bread, fruit juice, beer, etc. and in non-food industries such as ethanol production, textiles and detergents. The purpose of this study was to isolate and characterize the coal mine waste water bacteria as the amylase enzyme producer. Isolation of mine water bacteria isolate was done by scratch grate method using Nutrient Agar (NA) medium, obtained from 5 single colony then continued with identification using Gram stain, Capsic Dye, Spore Dye, and Biochemical Test. After identification followed by activity test of bacterial waste water of coal mine in yielding enzyme amylase using diffusion method of wells which then continued with iodine test. The results of this study indicate the five bacterial isolates are Gram positive bacteria, which have a capsule, and spores. Bacterial isolates LTB2 had the largest inhibitory zone diameter of 15.5 mm, isolate LTB1 of 15.0 mm, isolates LTB4 of 14.7 mm, isolates LTB3 of 14.3 and isolates LTB5 had the smallest inhibitory zone diameter of 13.1 mm. Where the greater the diameter of the inhibitory zone the more enzyme amylase produced. Keywords: Amylolytic bacteria, liquid-waste of coal
