Abstract
Black rice (Oryza sativa L.) contains flavonoid and tanin which have mechanism as antibacterial. The purpose of this study is to determine antibacterial activity of ethanolic extract, n-hexane, ethyl acetate, and water fraction from black rice against Staphylococcus aureus ATCC 25923. Black rice was extracted by maceration using 96% ethanol solvent and fractionated using n-hexane, ethyl acetate, and water solvents. The results of extracts and fractions were tested for antibacterial activity using 10%, 15% and 20% concentrations. Negative control used 7% DMSO and Ciprofloxaxin as a positive control. Antibacterial activity determined by using disc diffusion and dilution method. The results of the antibacterial activity showed that ethanol extract, nhexane, ethyl acetate, and water fraction from black rice had antibacterial activity. Ethyl acetate fraction with 20% concentration is the most active fraction with an average diameter inhibition zone of 13.06 mm and 5% of MBC. Thin Layer Chromatography results showed that ethyl acetate fraction has flavonoid, tannin and triterpenoid compounds. Kata kunci: antibacterial, black rice (Oryza sativa L. Indica), fractination
