Abstract
Stevia leaves contain some chemical compounds serving as antibacterial agent, alkaloid, steroid, tannin, saponin, and flavonoid. This research aimed to find out the antibacterial activity of ethanol extract, n-hexane, ethyl acetate, and water fraction of stevia leaves, to find out which one has most effective antibavterial activity of ethanol extract, n-hexane, ethyl acetate, and water fraction of stevia leaves, and to find out the MIC and MBC of ethanol extract, n-hexane, ethyl acetate, and water fraction of stevia leaves on P. aeruginosa ATCC 27853. The antibacterial test in this study was conducted using diffusion and dilution methods. The concentrations of extract and fraction in diffusion method were 12,5, 25 and 50%. Ethyl acetate fraction concentration in dilution method were 50; 25; 12,5; 6,25; 3,125; 1,562; 0,781; 0,390; 0,195 dan 0,0975%. Ethyl acetate fraction of stevia leaves was identified for its chemical content using TLC method. Ethanol extract, n-hexane, ethyl acetate, and water fraction of stevia leaves has antibacterial activity. Ethyl acetate fraction 50% was the most effective one with inhibiting zone diameter of 14,25 mm. MIC cannot be determined, and MBC value of ethyl acetate fraction on P. aeruginosa ATCC 27853 is 6,25%. Keywords : stevia leaves, ethyl acetate fraction, P. aeruginosa ATCC 27853, MBC
