Abstract

Bay leaves are leaves that are efficacious for treating diarrhea. The chemical content of bay leaves is alkaloids, flavonoids, saponins, and tannins. These compounds have activities as antibacterial. This study was conducted to determine the antibacterial activity of ethanol extract, n-hexane fraction, ethyl acetate and bay leaf water to Escherichia coli ATCC 25922, find out the most active fraction and find out the Minimum Inhibitory Concentration (MIC) and Minimum Killer Concentration (KBM) from the extraction fraction bay leaf ethanol. Salam leaf powder was macerated using 70% ethanol then concentrated followed by fractionation using n-hexane, ethyl acetate and water. The results of extraction and fractionation were tested for antibacterial activity against Escherichia coli ATCC 25922 using the diffusion method, to find out the most active fraction. The concentration of extracts and fractions used for diffusion tests were 50%, 25% and 12.5%. The diffusion test results were continued by dilution tests, to determine KHM and KBM with a concentration series of 50%, 25%, 12.5%, 6.25%, 3.125%, 1.5625%, 0.7813%, 0.3906%, 0 , 1959%, and 0.0977%. Diffusion data obtained were analyzed by ANOVA. The results of the diffusion test showed that the extract, n-hexane, ethyl acetate and water fraction had antibacterial activity against Escherichia coli ATCC 25922. The most active fraction was ethyl acetate fraction with a KBM value of 25%. Keywords: bay leaves (Syzygium polyanthum [Wight] Walp), Antibacterial, diffusion, dilution, Escherichia coli ATCC 25922.
