Abstract

Urinary Tract Infection (UTI) is one of the highest prevalence disease in the world. One of causative agents of UTI is Escherichia coli. Macroalgae such as latoh (Caulerpa racemosa) contain compounds which have antibacterial activity such as flavonoids, alkaloids, terpenoids, tannins and saponins. The aim of this study was to determine the antibacterial activity of extract and n-hexane, ethyl acetate, water fractions against Escherichia coli ATCC 25922. The extract was obtained by maceration using ethanol 96% continued with fractionation using n-hexane, ethyl acetate and water. The assay was conducted using disc diffusion and dilution method. Screening antibacterial activity of extracts and n-hexane, ethyl acetate, water fractions through diffusion method by measuring the diameter of the inhibitory zone. The most effective concentration of n-hexane fraction is continued with dilution method to determine MIC and MBC value. The diameter of the inhibition zone was analyzed by using One Way ANOVA. The results showed that extract and n-hexane, ethyl acetate, water fractions had antibacterial activity. The diffusion method in the n-hexane fraction of 30% concentration had the greatest antibacterial activity against Escherichia coli ATCC 25922 with a diameter of inhibition zone of 18,5 mm. The dilution method of n-hexane fraction showed the MIC value cannot be determined and MBC value of concentration 60%. Keywords: diffusion, dilution, Escherichia coli, latoh
