Abstract

The main objective of this study was to improve solubility and know the character of Loratadin (LOR) after characterization. Loratadin (LOR) is an antihistamine that is not soluble in water and is class II BCS so that particle modification is carried out through Spherical Crystallization using spherical agglomeration. In this study DMSO, Ethyl acetate, water and PVP (Polyvinyl Pirolidone) are used as good solvents, connecting fluids and polymers. The choice of this solvent depends on the inability of the solvent and the solubility of the drug in individual solvents. Therefore DMSO, ethyl acetate, water is chosen as a good solvent, poor connecting fluid and solvent. Loratadine is soluble in DMSO but not soluble in water. The formed agglomerates were characterized by looking at opic microscopy, X-ray diffraction (XRD), calculated% yield and solubility test. The results of the microscope showed that the surface of the pure crystal loratadine which had originally changed to a round with a magnification of 100x. The results of the SEM test obtained a particle size of 300 μm with 200x magnification. In the XRD test, it was concluded that there were changes in the diffractogram peaks of spherical loratadin crystals because there was PVP and other factors such as temperature and stirring. The yield produced is 92.21%. The results of the study of spherical crystal solubility were 7,50354mg / ml and pure drugs 3,57568 mg / ml which showed that after modification of the particle surface, spherical crystals of loratadine were able to increase the solubility of pure drugs.
