Abstract

Secang heartwood (Caesalpinia sappan L.) is a Fabaceae plant which has antibacterial activity. The chemical content of secang wood is flavonoids, alkaloids, tannins, and saponins. This study aims to determine the activity of ethanol extract, n-hexane, ethyl acetate, and water from secang (Caesalpinia sappan L.) as an antibacterial against Pseudomonas aeruginosa ATCC 27853 by looking at MIC and MBC from the most active samples. Antibacterial activity test of ethanol extract, n-hexane, ethyl acetate, and water fraction against Pseudomonas aeruginosa ATCC 27853 using diffusion and dilution methods. The extract concentration and fraction used for the 50% diffusion method; 25%; and 12.5%. The most active sample concentration used for the dilution method is 50%; 25%; 12.5%; 6.25%; 3.12%; 1.56%; 0.78%; 0.39%. The most active extracts and fractions were tested for chemical content using a tube test. Ethanol extract, n-hexane, ethyl acetate, and water fractions from secang heartwood have antibacterial activity against Pseudomonas aeruginosa ATCC 27853. The most active sample is a 50% ethanol extract with a inhibition zone diameter is 24,43 mm. The Minimum Bactericidal Concentration (MBC) of ethanol extract against Pseudomonas aeruginosa ATCC 27853 respectively was 3.12%; 6.25%; 12.5%; 25%; and 50%. The results of the identification of tube tests showed that ethanol extract and ethyl acetate fraction contained flavonoids, saponins, and tannins.
