Abstract

Chives leaves contain chemical compounds such as saponins, triterpenoids, tannins, flavonoids, and allicin. This research aimed to find out antibacterial activity of n-hexane, ethyl acetate, and water fractions of chives ethanol extract, to find out the most active extract or fraction and to find out Minimum Inhibiting Concentration and Minimum Killing Concentration of most active extract or fraction on Streptococcus mutans ATCC 25175. Chives leaf powder was extracted by maceration method with 70% ethanol. The extract obtained was fractionated with n-hexane, ethyl acetate, and water solvents. The fractionation results were tested for antibacterial activity against Streptococcus mutans ATCC 25175 using a diffusion method with a concentration of 50, 25, 12.5% to find out the most active fraction. The most active fraction was continued using the dilution method to determine the Minimum Inhibitory Concentration and Minimum Killer Concentration with concentrations of 50, 25, 12.5, 6.25, 3.125, 1.562, 0.781, 0.391, 0.195, and 0.098%. The results showed that the n-hexane, ethyl acetate, and water fractions of the ethanol extract of chives leaves had antibacterial activity against Streptococcus mutans ATCC 25175. The 50% concentration of ethyl acetate fraction had an average inhibition diameter of 16,67 mm. The most active fraction of Streptococcus mutans ATCC 25175 was the ethyl acetate fraction with an unknown Minimum Inhibitory Concentration and a Minimum Killer Concentration of 12.5%.
