Abstract

Soil is a good medium as a place to grow and develop diverse microorganisms. Enzymes derived from microorganisms are in great demand because they have many advantages such as fast growth, short life cycles, and high productivity. Enzymes that are widely known are amylase and cellulase. Amylase is an enzyme that can catalyze the breakdown of glycoside bonds from starch to simple sugars. Cellulase is an enzyme that is able to degrade cellulose through a catalyst process that works synergistically to release sugar. This study aims to isolate and identify Bacillus sp. bacteria obtained from tea plantation land which is capable of producing amylase and cellulase enzymes. This research is a qualitative descriptive study. The data obtained is presented descriptively which includes the identification of macroscopic, microscopic, qualitative test of amylase and cellulase activity. This study uses a diffusion method by means of bacteria inoculated on amylum media or CMC as a result of positive activity can be seen by the presence of a clear zone on the media. There were six bacterial isolates that had activity. Three isolates showed amylolytic activity as evidenced by the presence of clear zones around bacterial colonies on starch media. Three other bacterial isolates showed cellulolytic activity marked by the presence of clear zones in CMC media. The results of the identification showed that the six bacterial isolates were Bacillus sp. Keywords: amylase, cellulase, soil, Bacillus sp.
