Abstract

Breast cancer in women ranks the first cause of death compared to other cancers. Red ginger rhizome (Zingiber officinale Rosc Var. Rubrum) is empirically used as a traditional medicinal spice which is known to have anticancer activity. This study aims to determine the cytotoxic activity and the selectivity of the extract, ethyl acetate fraction and n-hexane fraction of red ginger rhizome against breast cancer cells T47D. Cytotoxic activity test of extract and fraction of red ginger rhizome were carried out by Microculture Tetrazolium Technique (MTT) method with concentration series (500; 250, 125; 62,5; 31,2; 15,6 and 7,8) µg/ml and positive control doxorubicin with a series of concentrations (1; 0.5; 0.25; 0.125; 0.06; and 0.03) µg/ml. The linear regression equation was made between log concentrations with % viability, used to calculate IC50. The selectivity of cytotoxic activity is known by the selectivity index equation, namely the IC50 value of vero cells compared to IC50 of T47D cells. The test results cytotoxic activity of extracts of ginger in red have IC50 of 87.794 µg/ml (<100 µg/ml) and IC50 ethyl acetate fraction of 52.991 µg/ml and the n-hexane fraction IC50 value of 44.730 µg/ml (˃ 30 µg/ml), showed moderate cytotoxic activity against T47D cells, whereas doxorubicin IC50 value lower than the red ginger rhizome is 1,532 ug/ml. Selectivity index extract, n-hexane, ethyl acetate, and water fraction on vero cells were obtained ˃ 3, while doxorubicin was 2,8 (˃3), indicating that doxorubicin can give normal cells toxicity.
