Abstract

Pletekan (Ruellia tuberosa L.) contains flavonoids, tannins, steroids and triterpenes which have antifungal activity. The purpose of this study was to determine the antifungal activity of n-hexane, ethyl acetate, and water fraction from ethanol extract leaves of pletekan against Candida albicans ATCC 10231. The results of extraction and fractionation method were carried out by antifungal activity test using a diffusion method with a concentrations of 40, 20, 10% and using a dilution method with concentrations of 40, 20, 10, 5, 2,5; 1,25;, 0,63;, 0,32 and 0,16%. The positive control used diffusion method was flucanozole 0,2% and the negative control used was DMSO 5%, while dilution method the positive control used was fungal suspension and the negative control used was most active fraction The results of the antifungal activity test using a diffusion method were analyzed with two way Analisis of Varian (ANOVA) method. The results showed that all fractions and ethanol extracts leaves of pletekan had antifungal activity against Candida albicans ATCC 10231. n-hexane fractions was the most effective fraction of antifungal activity, the results of identification of compounds by thin layer chromatography showed n-Hexane fractions containing triterpenes and steroids. The diameter of inhibition zone of nhexane fractions with a concentration of 40, 20 and 10% against Candida albicans ATCC 10231 were 18,67; 16,50 and 13,25 mm, respectively.
