Abstract
Carbopol and HPMC are gelling agent that are often used in preparation gel where the combination of the two of gelling agents is known to provide good physical quality and stability. Piroxicam is a Non-Steoidal Anti-Inflammatory Drugs (NSAID) group which in silico study is known to be used to cure diabetes wounds by inhibiting matrix metalloproteinase 9 (MMP 9) by forming hydrogen bonds in the residue of Proline 421, Alanine 189, Leucine 188 and Tyrosine 432. The purposes of this study are to know the physical quality and stability of piroxicam gel preparation, to know the effect of piroxicam gel on wound healing and to find the ratio of variations of gelling agent piroxicam gel preparation in wound healing. The preparation of piroxicam gel was made in a concentration of 0.5% with different base variations, formula I is base control; formula II contains a gelling agent with a concentration of 0.5% HPMC and 1.5% Carbopol; formula III with a concentration of HPMC 1% and Carbopol 1% and formula IV with a concentration of HPMC 1.5% and Carbopol 0.5% then physical quality is tested. Observation of healing time is done by observing the duration of healing of diabetic wounds on the rabbit's back which has been induced aloxan marked by the percent of wound closure. The data obtained were then analyzed using one way ANOVA. The results showed that all the formulas piroxicam gel have good physical quality and stability and the most effective diabetes wound healing activity is formula II. The effect of gelling agent concentration on piroxicam gel preparations tested in vivo to rabbit test animals is that the more the concentration of gelling agent, the active substance from the gel will be more difficult to release the active substance, the longer the healing time of wound.
