Abstract
Infectious disease is one of the problems in the health sector, including in Indonesia. Infection is characterized by tissue damage and followed by abscesses festering. Inappropriate use of antibiotics can cause resistance in the treatment of infections. Madagascar cockroach is a cockroach in the surrounding environment that is widely used as bird feed, Arwana fish and tarantula. The chemical content of Madagascar cockroaches includes the isoquinoline group, chromon derivatives, thiazine group, imidazole and sulfonamide pyrrole analogues, furanone and flavanone. Other studies mention that the Madagascar cockroach has antibacterial activity against Methicillin Sensitive Staphylococcus aureus, Methicillin Resistant Staphylococcus aureus and E. coli. This research was conducted to determine the antibacterial activity of Madagascar cockroaches and determine what class of compounds contained in Madagascar cockroach extracts and fractions against MRSA and P. aeruginosa ATCC 27853 by bioautographic methods. Madagaskar cockroach body extraction using maceration method used methanol solvent, followed by fractionation with n-heksan, ethyl acetate and water solvents. Identification of compound content by thin layer chromatography using mobile phase of butanol: acetic acid: water (4: 1: 5) and stationary silica gel GF254 phase, then preparative thin layer chromatography (PTLC) was carried out on the ethyl acetate fraction. Antibacterial activity testing was carried out by the diffusion method with the concentrations of extracts and fractions used were 60, 40, 20 ppm followed by TLC bioautography. The results showed that the Madagascar cockroach extract, fraction and isolate had antibacterial activity against P. aeruginosa ATCC 27853 and had no activity against MRSA. The diffusion method of ethyl acetate fraction at concentration of 60, 40, 20 ppm gave inhibitory activity of 17mm, 16,36mm and 15,33mm. The 60 ppm ethyl acetate fraction is the most active fraction. Identification TLC of ethyl acetate fraction was suspected to be a class flavonoid compounds. Bioautographic TLC test of ethyl acetate fraction obtained Rf 0,69 has activity against P. aeruginosa ATCC 27853. Isolation by PTLC of ethyl acetate fraction result 3 bands. The results of the bioautographic TLC showed the presence of clear areas in isolates A and C and there was growth in isolate B against P. aeruginosa ATCC 27853.
