Abstract
Rimpang kunyit dan daging buah asam banyak digunakan sebagai bahan utama untuk pebuatan jamu kunyit asam, jamu kunyit asam merupakan jamu yang sering dikonsumsi oleh ibu hamil. Ekstrak kunyit asam telah terbukti mengandung bioaktif antioksidan golongan flavonoid. Senyawa flavonoid dapat menghambat aktifitas-aktifitas enzim fosfodieterase yang menghidrolisis cAMP, peningkatan konsentrasi cAMP yang tinggi dapat menyebabkan hambatan akslerasi pertumbuhan. Tujuan dari penelitan ini adalah untuk mengetahui efek teratogenik pada fetus mencit setelah pemberian kombinasi ekstrak rimpang kunyit dan daging buah asam. Penelitian menggunakan 25 ekor mencit galur Swis-webster yang terbagi menjadi kelompok. Kelompok kontrol negatif diberikan CMC Na 0,5%, kelompok kontrol positif diberikan siklofosfamid 50 mg/kg bb mencit dan kelompok perlakuan I, II, III, diberikan 682,5; 1365; dan 2730 mg/kg bb mencit per hari. Kombinasi ekstrak rimpang kunyit dan ekstrak daging asam diberikan pada hari ke 6-15 (masa organogenesis), hari ke 18 mencit dilaparatomi dan diambil fetus mencit. Fetus mencit diawetkan dengan larutan Boiun’s lalu diamati kelainan-kelainan gross morfologi dan biometrika secara makroskopis. Hasil penelitian didapatkan kombinasi ekstrak rimpang kunyit dan daging buah asam pada dosis 2 (1.365 mg/kg bb) dan dosis 3 (2.730 mg/kg bb) dapat menyebabkan teratogenik. Dosis 2 mengalami kelainan berupa hemoragi, dosis 3 mengalami kelainan berupa resorbsi, hemoragi, penurunan berat dan panjang badan, dan ekor keriting. Kata kunci : Uji teratogenik, kombinasi ekstrak rimpang kunyit dan ekstrak daging buah asam Turmeric rhizome and tamarind fruit meat is widely used as the main ingredient for the production of tamarind turmeric, tamarind turmeric is a herbal medicine that is often consumed by pregnant women. Turmeric rhizome and tamarind fruit meat extract has been shown to contain bioactive antioxidant class of flavonoids. flavonoid compounds can inhibit the activities of the enzyme phosphodieterase that hydrolyzes cAMP, an increase in high cAMP concentrations can cause inhibition of growth acceleration. The purpose of this research is to determine the teratogenic effects on the fetus of mice after administration of a combination of turmeric rhizome extract and sour fruit flesh. The study used 25 Swiss-webster mice divided into groups. The negatif control group was given 0.5% CMC Na, the positive control group was given cyclophosphamide 50 mg/kg bw mice and the treatment groups I, II, III, were given 682.5; 1365; 2730 mg/kg bw mice per day. The combination of turmeric rhizome(Curcuma domestica) extract and tamarind (Tamarindus indica L) extract was given on days 6-15 (during organogenesis), on the 18th day, the rat was dilaparatomi and the fetus was taken. The fetus of mice was preserved with boiun's solution then macroscopically observed grosss morphological and biometrics abnormalities. The results showed a combination of turmeric rhizome (Curcuma domestica) extract and tamarind fruit (Tamarindus indica L) extract at a dose of 2 (1,365 mg / kg bw) and dose 3 (2,730 mg / kg bw) can cause teratogenic. Dose 2 has an abnormality in the form of hemorrhage, dose 3 has an abnormality in the form of resorption, hemorrhage, weight loss and body length, and curly tail.
