Abstract

Obesitas adalah penumpukan lemak yang berlebihan akibat ketidak seimbangan asupan energi dengan energi yang digunakan dalam waktu yang lama. Daun kelor tanaman obat herbal yang telah banyak digunakan salah satunya sebagai antiobesitas. Kandungan flavonoid, saponin, tanin dan steroid dalam daun kelor memiliki aktivitas sebagai penurun berat badan. Tujuan penelitian ini adalah untuk mengetahui aktivitas ekstrak dan fraksi daun kelor sebagai penurun berat badan pada tikus putih jantan galur Sprague dawley yang diinduksi pakan tinggi lemak. Daun kelor diekstraksi dengan metode maserasi menggunakan pelarut etanol 70%. Di fraksinasi dengan pelarut n-heksana, etil asetat dan air. Uji aktivitas penurun berat badan menggunakan 35 tikus putih dikelompokkan menjadi: kontrol normal, kontrol positif obeslim® 2,16 mg/200 g BB, kontrol negatif Na-CMC 0,5%, ekstrak daun kelor 125mg/Kg BB, fraksi n-heksana 3,54 mg/200 g BB, fraksi etil asetat 2,23 mg/200 g BB dan fraksi air 15,23 mg/200 g BB. Data yang diperoleh dianalisis menggunakan uji statistik Shapiro wilk, uji One Way ANOVA dan uji Post Hoc yaitu Tukey. Hasil penelitian menunjukkan fraksi n-heksana, fraksi etil asetat dan fraksi air dari ekstrak daun kelor mempunyai aktivitas sebagai penurun berat badan dan lemak abdominal. Fraksi air 15,23 mg/200 g BB memiliki nilai persentase penurunan berat badan sebesar 29,2% dan berat lemak abdominal 1,5742 gram mendekati nilai kontrol positif. Saponin, tanin, steroid dan flavonoid merupakan golongan senyawa yang memiliki aktivitas dalam menurunkan berat badan pada tikus putih jantan galur Sprague dawley yang diinduksi pakan tinggi lemak. Obesity is an excessive accumulation of fat due to the imbalances of energy intake with energy used for a long time. Leaf kelor herb medicinal plants that have been widely used one of them as antiobesity. The content of flavonoids, saponin, tannins and steroids in the leaves of kelor has activity as a weight loss. The purpose of this study was to find out the activity of extracts and the fractions of the leaves as weight-loss in the white rats of Sprague -induced high-fat deft strains. The kelor leaves are extracted by the maceration method using the 70% ethanol solvent. At fractionation with the solvent n-hexane, Ethyl acetate and water. Test weight-loss activity using 35 white mice grouped into: normal control, obesity positive control obeslim® 2.16 mg/200 g BB, negative control Na-CMC 0.5%, kelor leaf extract 125 mg/Kg BB, n-hexane fraction 3.54 mg/200 g BB, Ethyl acetate fraction 2.23 mg/200 g BB and water fraction 15.23 mg/200 g BB. The Data obtained is analyzed using Shapiro wilkstatistical Test, One Way ANOVA test and Post Hoc test is Tukey. The results showed n-hexane fraction, ethyl acetate fraction and water fraction of kelor leaf extracts having decreased body weight and abdominal fat. Water fraction 15.23 mg/200 gram BB has a weight loss percentage value of 29.2% and the abdominal fat weight of 1.5742 grams approaches the value of positive control. Saponins, tannins, steroids and flavonoids are a group of compounds that have an activity in losing weight in the white rats of Sprague dawley, which are induced by high fat feed.
