Abstract
Proteolytic bacteria are microorganisms with the ability to produce protease enzymes. Protease enzymes have been applied extensively in industry to health. This research is aim to know an optimization for incubation times of protease enzyme activity produced by the Staphylococccus aureus and Bacillus subtilis bacteria that produce higher protease enzyme activity. Many steps is taken in this research to identify bacteria, bacteria fermentation, and protease enzyme activities as well as to Staphylococccus aureus and Bacillus subtilis measuring the diameter of the clear zone formed in the milk skim media to achieve during the day's incubation process. The data acquired is analysed in a descriptive Analysis of Variance (ANOVA). Research indicates optimum enzyme protease activity produced from Staphylococccus aureus fermentation is 2.49 cm on the second day and optimum protease enzyme activities produced from Bacillus subtilis fermentation are 1.96 cm (6 in.) on the second day. Whereas the optimum enzyme protease activity produced by Staphylococccus aureus isolate is 2.12 cm (2.12 in.) on the third day and the optimum enzyme protease activity obtained from Bacillus subtilis isolate is 1.39 cm (1.39 cm) on the third day.
