Abstract

Sesbania grandiflora or Red Agathi known as vegetables especially for the people in Bulungcangkring village, Jekulo, Kudus. People belive that Red Agathi can be used as lactation booster ( lactagogue) for breastfeeding mother. This study aimed to determine the activity of lactagogue and the variation of the effective dose of Red Agathi ethanol extract on increasing weight of the rat pups and the histopathology of mammary glands lactacing rats. This research was conducted experimentally by using Parallel Group Post Test Only Design method. The samples used were 30 female lactacing rats which divided into 6 group. The first group as normal control. The second group as negative control (CMC Na 0.5%). The third group as positive control ( smooth lactacing, dose in the amount of 18 mg/kg rat’s weight). The fourth, fifth, and sixth group as Red Agathi ethanol extract test with dosage variation in the amount of 250 mg/kg rat’s weight, 500 mg/kg rat’s weight, and 1000 mg/ kg rat’s weight. The treatment were given for 14 days, by weighing rat pups before and after suckle everyday. The first weigh was done on 08.30 am (W1). After that, separate them from the female rats for 4 hours to 12.30 pm (W2) and then bring them back with the female rats on 13.30 pm (W3). After that, calculate the average of weight gain daily with this formula [(W3-W2) + (W2-W1)/4]. Furthermore, the data obtain were tested using One Way Anova analysis and continue with LSD test. The research result revealed that the administration of ethanol extract of Red Agathi give the impact on increasing weight of new born rats and the hispatology of mammary glands lactacing rats. Extract dose in the amount of 1000 mg/kg rat’s weight that given every day for 14 days treatment giving significant impact in increasing weight of rat pups and histopathology of mammary glands lactacing rats seen from the size of the diameter and number of alveoli glands. The extract dose in the amount of 1000 mg/kg rat’s weight was comparable to the positive control ( smooth lactacing, dose in the amount of 18 mg/kg rat’s weight) compare with the other doses and control that being used.
