Abstract

Cervical cancer is a disease that affects many women and the main causes of death in women due to Human Papilloma Virus (HPV). Therapeutic threatments that that can be done is chemotherapy, surgery, radiation, and immunotherapy, but these treatments can cause side effects. The existence of these side effects, this study was conducted, the aim was of knowing the cytotoxic activity of mangosteen leaf extract and fraction against HeLa cells and to determine the cytotoxic activity of the most potent mangosteen leaf fraction. In this study, the remaceration extraction methode was used by immersing the material using an appropriate solvent and fractionation to separate the components of mixture derived from the extract in order to obtain the main compound based on differences in polarity. The method used to test this cytotoxic activity is the MTT assay. The principle of the MTT method is that there is a reduction of the yellow tetrazolium salt which will produce purple formazan crystals. Then to see the cytotoxic activity is to determine the IC50 value. The results of the cytotoxicity test of mangosteen leaf extract and fraction against HeLa cells, namely ethanol extract, n-hexane fraction, ethyl acetate fraction, and water fraction had IC50 results of 194,536, 131,826, 112,668, and 82,224 g/mL. From the IC50 results, all samples are quite active, but the most potent is the water fraction with the smallest value. The water fraction contain compounds flavonoid, polyphenols, tannins, and saponins. . Keywords: Garcinia mangostana L., cervical cancer, HeLa cells, cytotoxic.
