Abstract

Depressive disorder is a serious medical disorder that has disorders of the neurotransmitters norepinephrine, serotonin and dopamine. Depression causes different behaviors in test animals, such as silence or immobility, decreased hygiene or grooming behavior. Depression also causes an overproduction of cortisol, leading to high blood glucose levels. The brazilin compound purified from secang wood (Caesalpinia sappan L.) has the potential to be an antidepressant. The purpose of this study was to determine the antidepressant activity of secang wood ethanol extract and its effect on blood glucose levels in mice induced by forced swim tests. Male mice were adapted for 7 days and then depressed using the forced swim test method for 11 days with a duration of 5 minutes. The doses used in this study were 10.69; 21.38 and 42.74 mg/KgBB. The positive control used was fluoxetin 0.052 mg/20 gBB mice and the negative control used was 0.5% Na CMC. Mice were measured for imobility time using the forced swim test method, the duration of grooming with a sucrose splash test and blood glucose levels using a glucometer. The results showed that the ethanol extract of secang wood with a dose of 10,69; 21,38 and 42,76 mg /KgBB were able to significantly reduce time immobility by 42,56; 44,43 and 50,03% and the decrease in blood sugar levels respectively 12,43; 16,84 and 24,71%. The duration of the show shows negative immobility time values. The conclusion of this study is that the induction of FST leads to depression, increased IT, decreased treatment and increased blood glucose levels. The effective dose for reducing depression was at a dose of 10,69 mg / KgBB, while the effective dose for reducing blood glucose levels was at a dose of 4,76 mg / KgBB.
