Abstract

Cervical cancer is the second most common type of cancer suffered by women after breast cancer and is the highest cause of death in Indonesia. Alternative cancer treatment is currently back to natural ingredients. This study aims to determine the effect of extract and fraction of berenuk leaves on cytotoxic activity with parameter IC50 and to determine the components of compounds that have the potential to cervical cancer cells (HeLa). Extraction of berenuk leaves was carried out by maceration method followed by liquid-liquid fractionation with n-hexane, ethyl acetate and characterization to control the quality of extracts and fractions. Extracts and fractions were tested for their effects on HeLa cells using the MTT method with a concentration series of 250; 125; 62.5; 31.25; 15.75; 7.81; 3.75; 1.875 g/mL. Data analysis was carried out by calculating the parameter IC50 value and percent cell viability. The test results on the n-hexane and ethyl acetate fractions showed moderate active cytotoxic activity against cervical cancer cells (HeLa) with IC50 values of 83.56 g/mL and 74.13 g/mL, while the ethanol extract of berenuk leaves didn’t have significant cytotoxic activity potent with IC50 112.46 g/mL. Based on this, the presence of flavonoid, alkaloids, tannins, triterpenoids compounds in ethyl acetate fraction is expected to produce a cytotoxic effect on cervical cancer cells (HeLa).
