Abstract

The enzyme is a biomolecular that can function as a catalyst in some reactions. There are two types of enzymes based on their role, namely extracellular enzymes and intracellular enzymes. Extracellular enzymes are enzymes that work the outside of a cell, while intracellular enzymes work the inside of a cell. Some of the most needed extracellular enzymes are proteases and amylase. Some bacteria of the genus Bacillus one of them are Bacillus cereus, can produce extracellular enzymes such as protease and amylase. This research aims to determine the proteases and amylase enzyme activity in Bacillus cereus bacteria isolated from mangrove forest water. Enzyme activity was tested after precipitating the enzyme using ammonium sulfate with a concentration of 50% for each enzyme. There are differences between enzyme activity testing of protease and amylase. Bergmeyer and Grassl’s method is used for protease activity tests by calculating the absorbance of the blank, tyrosine standard, and enzyme sample with 1%; 1,5%; 2% of casein substrate concentration. Bailey’s method is used for amylase activity tests exerting DNS reagent and 1%; 1,25%; 1,5% of amylum substrate concentration and then enzyme activity calculated by substituting the absorbance of sample to glucose standard curve equation. Bacillus cereus bacteria have extracellular protease enzyme activity characterized by the formation of clear zones in sma media (Skim Milk Agar), while for amylase there is a clear zone around the colony after being edgydine in na (Nutrient Agar) media mixed with amyloid. The best concentration of substrates for protease enzyme activity is 2% (628.8 x 10-3 U/mL), while the best concentration of substrates for amylase enzyme activity is 1.5% (61.2 x 10-3 U/mL).
