Abstract
Cervical cancer or cervical cancer is the biggest cause of death in women, this is caused by a virus in the form of the Human Papilloma Virus (HPV). Basil leaves (Ocimum basilicum L) have the potential to have cytotoxic activity against HeLa cell cultures. The aim of this cytotoxic test was to determine the cytotoxic activity and IC50 value that resulted in 50% cell death of the ethanol extract, chloroform fraction and ethyl acetate fraction of basil leaves on HeLa cell culture and to determine the class of compounds that play a role in cytotoxic activity. Cytotoxic activity test of ethanol extract, chloroform fraction and ethyl acetate fraction of basil leaves used the MTT method. The principle of the MTT method is to reduce the soluble yellow salt to form an insoluble blue-purple formazan precipitate. Cytotoxic testing of the chloroform fraction was carried out at concentrations of 250 g/mL, 125 g/mL, 62.5 g/mL, 31.25 g/mL, 15.625 g/mL, 7.81 g/mL, 3.78 g/mL and 1.875 g/mL. Data analysis using a Microplate reader with a wavelength of 595 nm. The results of the cytotoxic test in this study showed that the IC50 value of the ethanol extract was 168.267 g/mL, the chloroform fraction was 22.439 g/mL and the ethyl acetate fraction was 36.308 g/mL. The most toxic IC50 value was the chloroform fraction and the one that had no cytotoxic activity was the ethanol extract. Compounds in the chloroform fraction that affect the inhibition of cytotoxic activity are flavonoids, alkaloids, tannins and steroids/triterpenoids.
