Abstract

Breast cancer is one of the cancers that always increases every year. Various methods used today still have shortcomings such as side effects to the occurrence of metastases, therefore new drugs from herbal plants are needed which are considered better than other healing methods. The content of compounds contained in the bark of faloak (Sterculia quadrifida R.Br) namely flavonoids, alkaloids, and anthraquinones is estimated to have cytotoxic activity that can be used as anticancer. This study aimed to examine the cytotoxic activity in the extract and fraction against MCF-7 cells, as well as to determine the class of compounds contained in the active fraction of faloak stem bark. Preparation of faloak bark powder extract using the remaceration method with 96% ethanol as solvent. The extract was then fractionated using a liquid-liquid extraction method using n-hexane, ethyl acetate, and water as solvents. The results of the extract and bark fraction of faloak were tested for cytotoxic activity in vitro against MCF-7 breast cancer cells using the MTT assay method by determining the IC50 value to see the toxic activity against MCF-7 cells. Cytotoxic activity test showed that the extract and bark fraction of faloak had cytotoxic activity within the active criteria known from the IC50 value of the extract, ethyl acetate fraction, water fraction and n-hexane fraction, respectively, namely 12.227 g/mL; 10.231 g/mL; 11,667 g/mL; and 16.863 g/mL and the ethyl acetate fraction which is the most potent fraction against cancer cells because it has the smallest IC50 value. Phytochemical screening test using TLC showed the presence of flavonoid, alkaloid and anthraquinone compounds contained in the ethyl acetate fraction.
