Abstract
Rosella flowers (Hibiscus sabdariffa L.) and binahong leaves (Anredera cardifolia (Ten) Steenis) are herbal plants that contain saponins, flavonoids, tannins, triterpenoids, steroids, and essential oils that have antibacterial effects. This study aims to test and prove the antibacterial activity of the combination of ethanol extract of rosella flowers and binahong leaves to compare the combination of the most effective comparison concentrations against Staphylococcus aureus. This study extracted rosella flowers (Hibiscus sabdariffa L.) and binahong leaves (Anredera cardifolia (Ten) Steenis) using the maceration method with 96% ethanol solvent. To determine the most effective concentration, bacteria were not grown with a concentration of 10% rosella flowers and 25% binahong leaves using the dilution method. This extract was prepared in a ratio of (1:1), (1:2) and (2:1) and then tested for antibacterial activity using the disc diffusion method and the results were analyzed using SPSS 21 statistics. The results of the comparison of the most effective concentrations of 2:1 were carried out combined effect by means of the paper tape method. The results showed that the combination test of rosella flower extract and binahong leaf had the activity of Staphylococus aureus ATCC 25923. The combination concentration of 2:1 was the most effective combination with an average inhibition zone diameter of 22.7 mm and gave a synergistic combination effect against Staphylococus aureus ATCC. 25923.
