Abstract

Breast cancer is one of the most dangerous diseases that attack the breast organs in women and one of the most common causes of death, with 18.1 million cases diagnosed. Cancer drugs currently have very dangerous side effects, so the discovery of natural therapy is needed. One of the plants used as anticancer is melinjo seeds (Gnetum gnemon L.). Melinjo seeds are plants that contain flavonoids, alkaloids, and steroids/terpenoids that can act as natural medicines. The purpose of this study was to determine the cytotoxic activity of the extract and fraction of melinjo seeds and the class of compounds in the melinjo seed fraction that had the most active cytotoxic effect on MCF-7 cells. This study used an experimental method. Melinjo seed powder was extracted by maceration method then the resulting thick extract was fractionated using a liquid-liquid extraction method with n-hexane, ethyl acetate, and water as solvents. Cytotoxic test using the MTT assay method where the resulting absorbance results will be analyzed using Microsoft Excel to get the IC50 value. Cytotoxic activity test showed that the extract and fraction of nhexane, ethyl acetate, and water in melinjo seeds had cytotoxic activity against MCF-7 breast cancer cells with IC50 values of 120,218, respectively; 9.77; 44.66; 29.030 g/mL with n-hexane as the most active fraction and a class of compounds that have potential as cytotoxic agents are steroid compounds Keywords : Gnetum gnemon L., melinjo seed extract, cytotoxic, apoptosis, MCF-7 cell
