Abstract

Thrombocyte calculation is a significant examination of various cases, either dealing with hemostasis or other cases that include diagnosis validation, evaluation of therapy result or history of a disease, prognosis determination and evaluation of the level of a disease. Thus, examining thrombocyte count has to consider pre-analytical, analytical and post-analytical steps. One of factors influencing platelet count is the use of anti-coagulant, in which sometimes the use of EDTA anti-coagulant can cause pseudo-thrombocytopenia case. Therefore, samples have to be re-taken by changing the anti-coagulant with Sodium Citrate (Na. Citrate). This study aims at investigating the comparison the results of thrombocyte (platelet) counts in blood smear anti-coagulated with EDTA and Sodium Citrate. This study applied comparative analytical design with cross-sectional approach. There were 30 samples used in this research. This research was carried out in the Clinical Pathology Laboratory of Regional Public Hospital (RSUD) of Karang Asem in Bali in February 2017. The examination of thrombocyte count was performed using the method proposed by Barbara Brown, namely estimation calculation. Data were analyzed using paired-sample t-test with p<0.05 (significant) and confidence interval of 95%. The results show that the average thrombocyte count in blood smear with EDTA anti-coagulant was 245.670/μL and with Na. Citrate anti-coagulant was 248.670/μL, with (p value) sig (2 - tailed) of 0.000<0.005. It concludes that there are significant differences of thrombocyte counts in blood smear with EDTA anti-coagulant and with Na. Citrate anti-coagulant. Keywords: Thrombocyte, EDTA, Na.
