Abstract
Acyclovir used in the treatment and prevention of infectious diseases caused by HSV and VHZ are included BCS Class III which has high solubility, but low permeability therefore modification of the nanometer sized preparations. The purpose of this research is to know the characteristics of nanoparticles and get the best formula based on the particle size, and to know whether the formula is selected can be made gel preparations that meet the good physical properties. In this research were used 5% powder of acyclovir as an active ingredient. Chitosan : tripolyphosphate F1 (0,1% : 0,2%), F2 (0,2% : 0,2%), F3 (0,3% : 0,1%). Acyclovir have been made nanometer use ionic gelation method, then formulated to gel form using HPMC as gelling agent. The result have been charactherized for particle size continued formula 1, 2, 3 is 1708 nm±491,43; 572,2 nm±491,43; 794,8 nm±491,43. Polydispersity index 0,769±0,026; 0,714±0,026; 0,712±0,026. Zeta potensial +25,6±3,226; +33,4±3,226; +30,6±3,226; entrapment efficiency 99,87%±0,03; 99,93%±0,03; 99,88%±0,03. The results of the characterization, formula 2 showed better results with concentration variations chitosan 0,2% and tripolyphosphate 0,2%. Formula 2 formulated to gel and result the best quality of gel with the result of one sample t-test of viscosity, adhesiveness, and distribution showed that there was no significant difference between prediction and the result of real test.
