Abstract

The wake-up plant (Coleus ambonicus L.) is known in the North Sumatra area as long as its leaves are often used as lactagogum. In addition to being used as lactagogum, this plant also contains many compounds, one of which is flavonoid compounds which are thought to provide a tonic effect. This study aims to determine the tonic effect as well as find out the effective dose of wake-up leaf ethanol extract which can have a tonic effect on male white mice. In this study, the method chosen to perform extraction was the maceration method, with the tonic rotarod test and natatory exhaustion test methods. The test used 25 mice test animals and then divided into 5 treatment groups. Group I (negative control) is CMC Na 0.5%, group II is (positive control) i.e. caffeine 13 mg/kgBB mice, group III, IV, and V successively wake-up leaf extract 30 mg/20 gBB mice, 60 mg/20 g BB mice, 120 mg/20 g BB mice. The data collected were in the form of fatigue time of mice before and after treatment from the rotarod test and natatory exhaustion methods. The results obtained were analyzed using shapirowilk to see the distribution of data. The results showed that the wake-up leaf extract had a tonic effect on male white mice with an effective dose of 120 mg / 20 gBB of mice. In the rotarod test method, the average increase in fatigue time is 25.2 seconds. The average increase in fatigue time for each group I, II, III, IV, V respectively was 9 ; 35.2 ; 13 ; 19.6 ; 25.2 seconds. Then in the natural exhaustion method, the average increase in fatigue time is 5.5 minutes. . The average increase in fatigue time for each group I, II, III, IV, V respectively was 1.55 ; 6,10 ; 3.17 ; 3.32 ; 5.5 minutes. Key words : Tonic, Wake-up leaves, Rotarod test, Natatory exhaustion
