Abstract
The tempuyung plant is a plant that is empirically useful in the health sector, one of which is as an antibacterial. Bacterial infections often occur in society, caused by Staphylococcus aureus which causes dermatitis or mass poisoning, for Escherichia coli bacteria can cause indigestion in humans. The content of active compounds in tempuyung plants has the potential as an antibacterial which is suspected to be a secondary metabolite compound. This study used three kinds of solvents with different polarity properties, including n-hexane, ethyl acetate, and 70% ethanol. The leaves of tempuyung in stratified maceration using three kinds of solvents aimed to see what extracts from solvents with polarity would be more effective when used for KHM and KBM testing against Staphylococcus aureus and Escherichia coli. The test was carried out by the method of diffusion and dilution, the diffusion method was carried out by scraping bacteria onto a petri dish containing the planting medium after which it was attached to the disc. The result of the diffusion method is used for the dilution method. The results of the study can be concluded that tempuyung extract with 70% ethanol solvent has antibacterial activity. The degree of polarity of the solvent affects the effectiveness of antibacterials. The results of the KHM and KBM tests showed that ethanol polarity levels were 70% more effective compared to other solvents. The concentration of 50% has good inhibitory power as an antibacterial and 12.5% as a minimum concentration of killing power for Escherichia coli bacteria.
