Abstract

HERTANTI, D.D., 2023. OPTIMIZATION OF PYROXICAM TABLET FLOATING FORMULATION WITH CHRONOTHERAPEUTIC DRUG DELIVERY SYSTEM. Chronotherapeutic system (CHT) is a specific place and time controlled drug release with a programmed delivery system. Piroxicam CHT floating tablet formulation is the right alternative to increase the comfort of use in geriatric patients by providing drug release when symptoms worsen so that when you wake up, pain is not felt. Piroxicam is a non-steroidal anti-inflammatory drug (NSAID) used especially by geriatrics for the treatment of rheumatoid arthritis (RA) and osteoarthritis (OA). This study aims to determine the optimal concentration and the effect of the combination of polymers HPMC K4M, HPMC K100M and Carrageenan on the physical properties and dissolution rate of piroxicam CHT floating tablets. Simplex lattice design method used to determine the effect and interaction of the physical properties tablet and dissolution speed of 7 formulas piroxicam floating tablets with combination of HPMC K4M, HPMC K100M and Carrageenan polymers. Tablets were compressed with a weight of 300 mg by direct compression method. Critical point parameters include flow time, water absorption, floating lag time, total floating time, and dissolution. Having obtained the optimum formula was compared between the experimental results with predicted results then were analyzed by t-test simplex one. Concentration of 800 mg and 199 mg (HPMC K4M : Carrageenan) is an optimum formula with the results of one test t-test simplex ρ>0.05 so that there is no significant difference between the experimental results and the predicted results. Carrageenan greatly influences the increase in floating lag time, friability and dissolution and decreases tablet hardness. Combination of HPMC K4M and Carrageenan the lower the friability and floating lag time and increase the tablet hardness and drug release
