Abstract
ROYHAN JURNERBOY, 2023, ANTIDEPRESSANT ACTIVITY TEST OF MARIGOLD FLOWER EXTRACT (Tagetes erecta L.) WITH ETHANOL SOLVENT IN MALE MICE (Mus musculus), THESIS, FACULTY OF PHARMACY, SETIA BUDI UNIVERSITY, SURAKARTA. Supervised by Dr. apt. Ika Purwidyaningrum, S.Farm., M.Sc and apt. Yane Dila Keswara, M.Sc. Depression is a psychological disorder in humans that can be overcome by using antidepressant drugs. The use of these antidepressant drugs can cause side effects and potential dependence. Marigold flower extract contains alkaloids, flavonoids, and quercetin compounds that are known to have antidepressant activity. This study aimed to evaluate whether ethanol extract from marigold flowers had antidepressant effects, as well as to determine the effective dose of the extract as an antidepressant in male white mice (Mus musculus). Marigold flower plant extract (Tagetes erecta L.) was obtained by maceration process, then antidepressant testing using the tail suspension test method by observing the immobility time and behavior of mice before and after treatment of male white mice with a negative control of CMCNa 0.5%, a positive control group of amitriptyline dose of 25mg, and a dose of marigold flower extract 12.5; 25; 50 mg/kg body weight. Immobility time and behavioral observations were conducted on days 7, 14, and 21. The measurement data were analyzed using the Shapiro-Wilk Test normality test and homogeneity test. After meeting the requirements, then proceed with the One way ANOVA test then the Tukey test. The results showed that ethanol extract of marigold flowers (Tagetes erecta L.) has activity as an antidepressant seen from immobility time with the tail suspension test method. The effective dose as an antidepressant is 50 mg / kg body weight from marigold flower extract. Yane Dila Keswara, M.Sc. Depresi merupakan suatu gangguan psikologis pada manusia yang dapat diatasi dengan menggunakan obat antidepresan. Penggunaan obat antidepresan ini dapat menimbulkan efek samping dan potensi ketergantungan. Ekstrak bunga marigold mengandung alkaloid, flavonoid, dan senyawa kuersetin yang dikenal memiliki aktivitas sebagai antidepresan. Penelitian ini bertujuan untuk mengevaluasi apakah ekstrak etanol dari bunga marigold memiliki efek antidepresan, serta untuk menentukan dosis efektif ekstrak tersebut sebagai antidepresan pada mencit putih jantan (Mus musculus). Ekstrak tanaman bunga marigold (Tagetes erecta L.) didapatkan dengan proses maserasi, kemudian dilakukan pengujian antidepresan menggunakan metode tail suspension test dengan mengamati immobility time dan perilaku dari mencit sebelum dan sesudah perlakuan mencit putih jantan dengan kontrol negatif CMCNa 0,5%, kelompok kontrol positif amitriptyline dosis 3,25 mg/kg BB mencit, dan dosis ekstrak bunga marigold 12,5 ; 25; 50 mg/kg BB mencit. Immobility time dan pengamatan perilaku dilakukan pada hari ke 7, 14, dan 21. Data hasil pengukuran dianalisis menggunakan uji normalitas Shapiro-Wilk Test dan uji homogenitas. Setelah memenuhi syarat, maka dilanjutkan dengan uji One way ANOVA kemudian uji Tukey. Hasil penelitian menunjukkan bahwa ekstrak etanol bunga marigold (Tagetes erecta L.) memiliki aktivitas sebagai antidepresan yang dilihat dari immobility time dengan metode tail suspension test. Dosis efektif sebagai antidepresan yaitu 50 mg/kg BB dari ekstrak bunga marigold. Pengamatan perilaku mencit selama uji coba didapatkan hasil kondisi mencit tampak normal, tidak ada efek bermakna yang dihasilkan oleh mencit selama perlakuan.
