Abstract
Research has been conducted to separate amino acids using thin layer chromatography (CLT) by determining the retention factor (Rf) value. Amino acids are important components of proteins that have various functions in the body. KLT is a simple chromatographic method that is often used to separate and identify chemical components in a mixture. In this study, KLT was used to separate four types of amino acids, namely histidine, tryptophan, glycine, and alanine. The KLT procedure includes silica plate preparation, sample bottling, chromatogram formation by elution of various solvents, and determination of the Rf value of each sample. The results showed that each amino acid sample has a different Rf value due to differences in the polarity of each amino acid to the stationary phase of the silica plate and the solvent used. However, the Rf values obtained are not in accordance with the theory because there are errors in the preparation of silica plates.
