Terhadap Jumlah Total Leukosit Pada Tikus Putih Yang di Infeksi Staphylococcus aureus. “Karya Tulis Ilmiah”. Program Studi D3 Analis Kesehatan, Fakultas Ilmu Kesehatan Universitas Setia Budi. Staphylococcus aureus merupakan bakteri patogen yang dapat menyebabkan terjadinya berbagai jenis infeksi dan digunakan sebagai antigen dalam penelitian ini. Salah satu yang berperan dalam melawan infeksi adalah leukosit. Jumlah total leukosit digunakan sebagai parameter untuk mengindikasikan adanya infeksi tubuh. Ekstrak daun kayu putih (Melaleuca leucadendron L.) digunakan sebagai immunomodulator untuk melindungi tubuh dari antigen. Penelitian ini bertujuan untuk mengetahui pengaruh ekstrak daun kayu putih terhadap jumlah total leukosit pada tikus putih yang di infeksi Staphylococcus aureus. Jenis penelitian ini menggunakan penelitian eksperimental dengan hewan coba tikus putih sebanyak 30 ekor yang dibagi menjadi 6 kelompok meliputi : kontrol normal, kontrol negatif, kontrol positif, kelompok perlakuan 1,2,3 yang masing-masing diberi ekstrak daun kayu putih dengan dosis 400,800 dan 1600mg/kgBB secara oral selama 21 hari. Pada semua kelompok perlakuan pada hari ke-15 kecuali kelompok kontrol normal diinjeksi bakteri Staphylococcus aureus secara Intraperitoneal. Data total leukosit dianalisis dengan menggunakan uji paired t-test dan uji one way annova. Hasil penelitian menunjukkan pada pemberian ekstrak daun kayu putih terdapat perbedaan secara nyata antara sebelum dan sesudah perlakuan. Pada uji one way annova tidak ada perbedaan secara nyata total leukosit pada semua perlakuan, akan tetapi pada perlakuan 800mg/kgBB cenderung menurunkan jumlah total leukosit yang terbesar. Faculty of Health. Setia Budi University. Staphylococcus aureus is a pathogenic bacterium that can cause various types of infections and is used as an antigen in this study. One that plays a role in fighting infection is leukocytes. The total leukocyte count is used as a parameter to indicate the presence of body infection. Eucalyptus leaf extract (Melaleuca leucadendron L.) is used as an immunomodulator to protect the body from antigens. This study aims to determine the effect of eucalyptus leaf extract on the total leukocyte count in white rats infected with Staphylococcus aureus. This type of research used experimental research with 30 white rats which were divided into 6 groups including: normal control, negative control, positive control, treatment groups 1, 2, 3, each of which was given eucalyptus leaf extract at a dose of 400,800 and 1600 mg/kg orally for 21 days. In all treatment groups on day 15 except the normal control group, Staphylococcus aureus was injected intraperitoneally. Total leukocyte data were analyzed using paired t-test and one way annova test. The results showed that in the administration of eucalyptus leaf extract there was a significant difference between before and after treatment. In the one way annova test there was no significant difference in the total leukocytes in all treatments, but the 800 mg/kgBW treatment tended to decrease the total leukocyte count the largest.
